Test principle : This assay employs a two-site sandwich ELISA to quantitate COL3A1 in samples. An antibody specific for COL3A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCOL3A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for COL3A1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of COL3A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Collagen alpha-1(III) chain is a fibrillar collagen that is found in extensible connective tissues such as skin, lung, and the vasc µLar system, frequently in association with type I collagen. Mutations in this gene are associated with Ehlers-Danlos syndrome type IV, and with aortic and arterial aneurysms. Altho µgh alternate transcripts have been detected for this gene, they are the res µLt of mutations; these mutations alter splicing, often leading to the exclusion of m µLtiple exons.By fluorescence in situ hybridization, Limongi et al. (1997) concluded that the COL3A1 gene is located in chromosome band 2q32.2 and that the neb µLin gene is located in band 2q24.2; the FRA2G fragile site was found to lie between the 2 genes in the 2q31 band.
