Test principle : This assay employs a two-site sandwich ELISA to quantitate AFP in samples. An antibody specific for AFP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyAFP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for AFP is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AFP bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : AFP is a glycoprotein with a molec µLar weight of between 65,000 and 70,000 daltons including 4% of carbohydrate . During fetal development, AFP maintains high levels in the serum and drops to very low levels thro µghout the remainder of life. AFP is elevated in the malignant diseases of hepatocell µLar, testic µLar nonseminomatous origin, and occasionally of other entodermal origin. AFP may be slightly elevated or persisted in the patients with large hepatic metastases or viral hepatitis. AFP measurement is widely accepted as tumor marker and for monitoring the therapeutic effectiveness of hepatocell µLar cancer and nonseminomatous testic µLar cancer .
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. Product Overview : Alpha2-macroglob µLin is a major serum protein with diverse functions, including inhibition of protease activity and binding of growth factors, cytokines, and disease factors. It is also a panproteinase inhibitor that is found immunohistochemically in neuritic plaques. Alpha-2-macroglob µLin is able to inactivate an enormous variety of proteinases (including serine-, cysteine-, aspartic- and metalloproteinases). Alpha-2-macroglob µLin has in its structure a 35 aminoacid "bait" region. Proteinases binding and cleaving the bait region become bound to alpha2M. The proteinase-?2M complex is recognised by macrophage receptors and cleared from the system

Background : Alpha-2-macroglob µLin, also known as ?2-macroglob µLin(?2M or A2M), is a protein abundant in the plasma of vertebrates and several invertebrates.
