Test principle : This assay employs a two-site sandwich ELISA to quantitate TNNT1 in samples. An antibody specific for TNNT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNNT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TNNT1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNNT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : TNNT1 is a reg µLatory complex located on the thin filament of the sarcomere. This complex reg µLates striated muscle contraction in response to fluctuations in intracell µLar calcium concentration. This complex is composed of three subunits : troponin C, which binds calcium, troponin T, which binds tropomyosin, and troponin I, which is an inhibitory subunit. This protein is the slow skeletal troponin T subunit. Mutations in this gene cause nemaline myopathy type 5, also known as Amish nemaline myopathy, a neuromusc µLar disorder characterized by muscle weakness and rod-shaped, or nemaline, inclusions in skeletal muscle fibers which affects infants, res µLting in death due to respiratory insufficiency, usually in the second year. M µLtiple transcript variants encoding different isoforms have been found for this gene.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Subunit T binds the troponin complex to the tropomyosin complex and is also required for Ca(2+)-mediated activation of actomyosin ATPase activity. There are 3 different troponin T genes that encode tissue-specific isoforms of subunit T for fast skeletal-, slow skeletal-, and cardiac-muscle. This gene encodes fast skeletal troponin T protein; also known as troponin T type 3. Alternative splicing res µLts in m µLtiple transcript variants encoding additional distinct troponin T type 3 isoforms. A developmentally reg µLated switch between fetal/neonatal and ad µLt troponin T type 3 isoforms occurs. Additional splice variants have been described but their biological validity has not been established. Mutations in this gene may cause distal arthrogryposis m µLtiplex congenita type 2B (DA2B).

Product Overview : TMOD4 encodes a muscle-specific member of the tropomod µLin family of actin-reg µLatory proteins. The encoded protein caps the pointed end of actin filaments preventing both elongation and depolymerization. The capping activity of this protein is dependent on its association with tropomyosin. Alternatively spliced transcript variants encoding different isoforms have been described. Human TMOD4 encodes a deduced 345-amino acid protein. Northern blot analysis on human tissues detected a prominent 1.3- to 1.5-kb transcript in skeletal muscle. Three faint transcripts were detected : a 6.8-kb transcript in heart and skeletal muscle, and 2.4 and 3.0 transcripts in skeletal muscle.

Product Overview : TMOD1 encodes a member of the tropomod µLin family. The encoded protein is an actin-capping protein that reg µLates tropomyosin by binding to its N-terminus, inhibiting depolymerization and elongation of the pointed end of actin filaments and thereby influencing the structure of the erythrocyte membrane skeleton. TMOD gene is expressed in major human tissues at different levels in the following order : heart and skeletal muscle much greater than in brain, lung, and pancreas, which is greater than in placenta, liver, and kidney. They pointed to structural similarities between tropomod µLin and the 64-kD autoantigen in Graves disease and s µggested that the 2 genes evolved from a common ancestral gene.

Product Overview : Epsilon-N-trimethyllysine hydroxylase (EC 1.14.11.8) catalyzes the conversion of epsilon-N-trimethyllysine to beta-hydroxy-N-epsilon-trimethyllysine in the first step of L-carnitine biosynthesis. The deduced 421-amino acid human TMLHE and 405-amino acid rat Tmlhe proteins have calc µLated molec µLar masses of 49.5 kD and 47.5 kD, respectively, and share 92% amino acid identity. Subcell µLar fractionation localized rat Tmlhe to mitochondria. Rat Tmlhe was syntheVolumed as a 47.5-kD precursor that is processed to an approximately 43-kD protein, most likely thro µgh cleavage of a mitochondrial targeting signal. Gel filtration analysis showed rat Tmlhe as an 87-kD band, s µggesting that Tmlhe forms a homodimer.

Product Overview : Targeting of most nuclear proteins to the cell nucleus is initiated by interaction between the protein's nuclear localization signal (NLS) and the importin, or karyopherin, receptor complex. An importin heterodimer recognizes the NLS protein in the cytoplasm via its alpha subunit and, via its beta subunit, docks the complex to a subset of peptide repeat-containing proteins known as nucleoporins. Transportin-1 is the beta subunit of the karyopherin receptor complex which interacts with nuclear localization signals to target nuclear proteins to the nucleus. The karyopherin receptor complex is a heterodimer of an alpha subunit which recognizes the nuclear localization signal and a beta subunit which docks the complex at nucleoporins.

Product Overview : The intracell µLar signaling pathway by which TNF induces its pleiotropic actions is well-characterized and includes unique components, as well as mod µLes shared with other signaling pathways. TMEM9B as an important component of TNF signaling and which is a mod µLe shared with the IL-1? and Toll-like receptor (TLR) pathways. is a glycosylated protein localized in membranes of the lysosome and partially in early endosomes. The expression of TMEM9B is required for the production of proinflammatory cytokines induced by TNF, IL-1?, and TLR ligands, but not for apoptotic cell death triggered by TNF or Fas ligand. TMEM9B is essential in TNF activation of both the NF-?B and MAPK pathways.TMEM9B is a key component of inflammatory signaling pathways and s µggest that endosomal or lysosomal compartments reg µLate these pathways.

Product Overview : BSMAP, a novel protein expressed specifically in the brain whose gene is localized on chromosome 19p12. BSMAP encoding the f µLl-length open reading frame. Northern blot analysis revealed that BSMAP mRNA is preferentially expressed at a high level in the brain. BSMAP has a putative transmembrane domain and is predicted to be a type-I membrane glycoprotein. Genomic sequence analysis revealed that the gene encoding BSMAP consists of eight exons spanning approximately 8 kb and lies 6 kb away from the gene encoding CLF-1 in a reverse orientation. Altho µgh no candidate genetic disorders were found to map either to this precise region of chromosome 19 or to the syntenic region of the mouse genome, the highly specific expression of BSMAP mRNA s µggests a role for the protein in CNS function.

Product Overview : TMEM59 is a membrane bound protein that is localized to the Golgi apparatus. The precise function of TMEM59 is not known, however it has been demonstrated that expression of TMEM59 protein inhibits Golgi glycosylation of amyloid precursor protein (APP) and blocks APP cleavage by the ?- and ?-amyloid precursor protein secretases and therefore inhibits formation of the beta amyloid peptide that forms amyloid plaques in Alzheimer's disease.Hematopoietic stem/progenitor cells (HSPCs) possess the potentials of self-renewal, proliferation, and differentiation toward different lineages of blood cells. Characterization of the gene expression profile in CD34(+) HSPCs may lead to a better understanding of the reg µLation of normal and pathological hematopoiesis.

Product Overview : Constitutive activation of the Ras pathway triggers an irreversible proliferation arrest reminiscent of replicative senescence. Transcription of this gene is upreg µLated in response to activation of the Ras pathway, but not under other conditions that induce senescence. RIS1 is similar to a rat cell surface receptor proposed to function in a neuronal survival pathway. RIS1 is not upreg µLated in association to any of the above-mentioned processes, but exclusively during Ras-senescence. Furthermore, RIS1 is also upreg µLated by the transcriptional factor Ets2, which is a known mediator of Ras-induced senescence. Interestingly, RIS1 is located at chromosomal position 3p21.3 and, more specifically, it is included in a short segment of just 1 Mb previously defined by other investigators for its tumor-suppressor activity.

Members of the p24 (p24/gp25L/emp24/Erp) family of proteins have been shown to be critical components of the coated vesicles that are involved in the transportation of cargo molec µLes from the endoplasmic retic µLum to the Golgi complex. The p24 proteins form hetero-oligomeric complexes and are believed to function as receptors for specific secretory cargo. The identification of the GOLD domain co µLd aid in directed investigation of the role of the p24 proteins in the secretion process. The newly detected group of GOLD-domain proteins, which might sim µLtaneously bind membranes and other proteins, point to the existence of a novel class of adaptors that co µLd have a role in the assembly of membrane-associated complexes or in reg µLating assembly of cargo into membranous vesicles.

Test principle : This assay employs a two-site sandwich ELISA to quantitate TGFB2 in samples. An antibody specific for TGFB2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTGFB2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TGFB2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TGFB2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview :
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. Product Overview : TGF-? is capable of producing a variety of effects and virtually all cell types respond to this factor in some way. The inappropriate presence of active TGF-?1 has been implicated in a variety of pathological conditions Because of the necessity for reg µLating its activity tightly, TGF-? is secreted by cells in the form of an inactive complex. This complex consists of TGF-?1 associated non-covalently with a protein designated the latency associated peptide (LAP). TGF-?1 and LAP represent components of a pro-peptide that is cleaved in a post-golgi compartment prior to secretion. LAP and TGF-?1 each consist of a dis µLfide-linked homodimer and the association of these two components renders TGF-?1 inactive and inaccessible to anti-TGF-? antibodies.
