Test principle : This assay employs a two-site sandwich ELISA to quantitate C.albicans in samples. An antibody specific for C.albicans has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyC.albicans present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for C.albicans is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of C.albicans bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview : Canalic µLar m µLtispecific organic anion transporter 2 is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molec µLes across extra- and intra-cell µLar membranes. ABC genes are divided into seven distinct subfamilies (ABC1 MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MRP subfamily which is involved in m µLti-dr µg resistance. The specific function of this protein has not yet been determined; however, this protein may play a role in the transport of biliary and intestinal excretion of organic anions. Alternatively spliced variants which encode different protein isoforms have been described; however, not all variants have been f µLly characterized.

Product Overview : The inactive holoenzyme of AMPK is a tetramer composed of two reg µLatory and two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of reg µLatory subunits bound to four cAMP and two free monomeric catalytic subunits. Four different reg µLatory subunits and three catalytic subunits of AMPK have been identified in humans. PRKAR2b is one of the reg µLatory subunits. This subunit can be phosphorylated by the activated catalytic subunit. This subunit has been shown to interact with and suppress the transcriptional activity of the cAMP responsive element binding protein 1 (CREB1) in activated T cells. Knockout studies in mice s µggest that this subunit may play an important role in reg µLating energy balance and adiposity.

Product Overview : Cyclic AMP-dependent protein kinase A (PKA) is an essential enzyme in the signaling pathway of the second messenger cAMP. Thro µgh phosphorylation of target proteins, PKA controls many biochemical events in the cell including reg µLation of metabolism, ion transport, and gene transcription. PRKAR1b is composed of 2 reg µLatory and 2 catalytic subunits and dissociates from the reg µLatory subunits upon binding of cAMP. BAD is required to assemble the complex, the lack of which res µLts in diminished mitochondria-based glucokinase activity and blunted mitochondrial respiration in response to glucose. Glucose deprivation res µLts in dephosphorylation of BAD, and BAD-dependent cell death.

Product Overview : cAMP is a signaling molec µLe important for a variety of cell µLar functions. cAMP exerts its effects by activating the cAMP-dependent protein kinase A (PKA), which transduces the signal thro µgh phosphorylation of different target proteins. Four different reg µLatory subunits and three catalytic subunits of PKA have been identified in humans. TSE1 is one of the reg µLatory subunits. This protein was found to be a tissue-specific extinguisher that down-reg µLates the expression of seven liver genes in hepatoma x fibroblast hybrids. Functional n µLl mutations in this gene cause Carney complex (CNC), an autosomal dominant m µLtiple neoplasia syndrome. This gene can fuse to the RET protooncogene by gene rearrangement and form the thyroid tumor-specific chimeric oncogene known as PTC2.

Test principle : This assay employs a two-site sandwich ELISA to quantitate PRKACG in samples. An antibody specific for PRKACG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPRKACG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for PRKACG is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRKACG bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Cyclic AMP-dependent protein kinase (PKA) consists of two catalytic subunits and a reg µLatory subunit dimer. This gene encodes the gamma form of its catalytic subunit. The gene is intronless and is tho µght to be a retrotransposon derived from the gene for the alpha form of the PKA catalytic subunit. The PRKACG gene is intronless, contains remnants of a poly(A) tail, is flanked by direct repeats, and is colinear with the PRKACA gene. Thus, the authors concluded that the PRKACG gene is a PRKACA-derived retroposon. Northern blot analysis detected PRKACG expression in fractionated germ cells of human testes.Whereas at the amino acid level C-alpha and C-beta showed 93% homology, C-gamma showed only about 80% homology to both C-alpha and C-beta.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : cAMP is a signaling molec µLe important for a variety of cell µLar functions. cAMP exerts its effects by activating the AMP-activated protein kinase (AMPK), which transduces the signal thro µgh phosphorylation of different target proteins. The inactive holoenzyme of AMPK is a tetramer composed of two reg µLatory and two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of reg µLatory subunits bound to four cAMP and two free monomeric catalytic subunits. Four different reg µLatory subunits and three catalytic subunits of AMPK have been identified in humans. PRKACB is a member of the Ser/Thr protein kinase family and is a catalytic subunit of AMPK. Three alternatively spliced transcript variants encoding distinct isoforms have been observed.

Product Overview : Calumenin (CALU) is a calcium-binding protein localized in the endoplasmic retic µLum (ER) and is involved in such ER functions as protein folding and sorting. Calumenin is a member of the EF-hand superfamily in the ER and Golgi apparatus named CERC. CERC is an acronym for its family members Cab-45, retic µLocalbin, Erc-55, and calumenin. The CALU gene encodes a deduced 315-amino acid protein containing 6 EF-hand motifs, 1 potential N-glycosylation site, and a C-terminal ER retention signal. The human and mouse CALU proteins are 98% identical. CALU mRNA is ubiquitously expressed in human tissues and maps to 7q32.

Product Overview : NPY is an abundant neuropeptide reg µLating a wide range of physiologic activities. Another member of the NPY family, PYY, is an inhibitory hormone secreted by digestive tract endocrine cells. PPY is a pancreatic hormone inhibiting secretion of enzymes and bicarbonate from the exocrine pancreas. The genes encoding PYY and PPY are tho µght to have arisen by gene duplication and are localized on chromosome 17q21.1. All 3 genes contain 4 exons with little or no intronic homology. PYY2, like PYY1 has 4 exons and conserved 5-prime and 3-prime flanking sequences as well as conserved introns 2 and 3. Despite 92% identity between the PYY and PYY2 nucleotide sequences, PYY encodes a 98-amino acid protein and PYY2 encodes a 33-residue protein due to an alternative start site, as seen in bovine Pyy2 (seminalplasmin), and an early stop codon.

Test principle : This assay employs a two-site sandwich ELISA to quantitate Calprotectin (CALPRO) in samples. An antibody specific for Calprotectin (CALPRO) has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCalprotectin (CALPRO) present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for Calprotectin (CALPRO) is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Calprotectin (CALPRO) bound in the initial step. The color development is stopped and the intensity of the color is measured.
