Test principle : This assay employs a two-site sandwich ELISA to quantitate TIPIN in samples. An antibody specific for TIPIN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTIPIN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TIPIN is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TIPIN bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : TIPIN associated with the components of the replicative helicase in human cell lines and protected cells against genotoxic agents. TIPIN was required for efficient cell cycle arrest in response to DNA damage, and depletion of TIPIN rendered cells sensitive to ionizing radiation as well as replication stress.It shares 72% amino acid identity with mouse Tipin, with the greatest divergence in the C termini. coexpression of Tim and Tipin in early embryonic and extraembryonic tissues, altho µgh Tipin was more widely expressed. After embryonic day 14.5, both Tim and Tipin were abundantly expressed in thymus, liver, gastrointestinal tract, and ventric µLar zone of brain, and Tipin was also expressed in lung. In ad µLt mouse brain, Tipin was expressed in thalamic and cortical regions, with lower expression in suprachiasmatic nuclei.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
