Test principle : This assay employs a two-site sandwich ELISA to quantitate CLU in samples. An antibody specific for CLU has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCLU present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for CLU is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CLU bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Clusterin (apolipoprotein J) is a 75 - 80 kDa dis µLfide-linked heterodimeric protein associated with the clearance of cell µLar debris and apoptosis. This protein has several Aliases : dimeric acidic glycoprotein (DAG protein), testosterone repressed prostate message-2 (TRPM-2), s µLfated glycoprotein-2 (SGP-2) and complement lysis inhibitor (CLI).This protein was first identified in 1988 in seminal fluid.In humans the gene is encoded on chromosome 8 (8p21) and is highly conserved between species (70-80% homology).It is expressed in most mammalian tissues and can be found in blood plasma, milk, urine, cerebrospinal fluid and semen. A number of proteins have been found to affect its expression including Egr-1 members from the AP-1 complex, HSF1/2, Cdx1/2 and B-Myb.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation.
