Test principle : This assay employs a two-site sandwich ELISA to quantitate DKK3 in samples. An antibody specific for DKK3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDKK3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for DKK3 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DKK3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : DKK3 is a protein that is a member of the dickkopf family. The secreted protein contains two cysteine rich regions and is involved in embryonic development thro µgh its interactions with the Wnt signaling pathway. The expression of this gene is decreased in a variety of cancer cell lines and it may function as a tumor suppressor gene. Alternative splicing res µLts in m µLtiple transcript variants encoding the same protein.DKK3 has 4 potential N-glycosylation sites and possesses an extended N-terminal domain preceding the first cys domain as well as an extended C-terminal region following the second cys domain. All DKKs have several potential sites for cleavage by furin-type proteases. Northern blot analysis revealed wide expression of a 2.5-kb DKK3 transcript, with highest expression in heart, brain, and spinal cord.
