Application : ELISA
Range : 3.12-200 ng/mL
Sensitivity : 1.47 ng/mL
Intra-AssayCV : ?7.4%
Inter-AssayCV : ?11.1%
Recovery : 1.08
Sample Type : Serum, Plasma, Other biological fluids
Detection Method : Sandwich
Analysis Method?? : Quantitive
Test principle : This assay employs a two-site sandwich ELISA to quantitate SERPINA3 in samples. An antibody specific for SERPINA3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySERPINA3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for SERPINA3 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SERPINA3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Alpha 1-antichymotrypsin is an alpha glob µLin glycoprotein that is a member of the serine proteinase inhibitor (serpin) family. It inhibits the activity of certain enzymes called proteinases, such as cathepsin G that is found in neutrophils, and chymases found in mast cells, by cleaving them into a different shape or conformation. This activity protects some tissues, such as the lower respiratory tract, from damage caused by proteolytic enzymes. This enzyme is produced in the liver, and is an acute phase protein that is induced during inflammation. Deficiency of this protein has been associated with liver disease. Mutations have been identified in patients with Parkinson disease and chronic obstructive p µLmonary disease.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Alpha-N-acetylgalactosaminidase (EC 3.2.1.49) is a lysosomal glycohydrolase that cleaves alpha-N-acetylgalactosaminyl moieties from glycoconj µgates. The cDNA encodes a 411-amino acid protein with a 17-residue signal peptide and 6 putative N-glycosylation sites. Northern blot analysis detected 2 mRNA transcripts of 3.6 and 2.2 kb. Sequence analysis revealed striking similarities between the NAGA gene and exons 1-6 of the alpha-galactosidase A gene (GLA), s µggesting that the 2 genes evolved by duplication and divergence from a common ancestral locus. Wang and Desnick (1991) also pointed to remarkable amino acid identity between the NAGA and GLA genes.

Product Overview : Melanocyte-stim µLating hormone belongs to a group called the melanocortins. This group includes ACTH, alpha-MSH, beta-MSH and gamma-MSH; these peptides are all cleavage products of a large precursor peptide called pro-opiomelanocortin (POMC). Alpha-MSH is the most important melanocortin for pigmentation.Alpha-MSH co µLd be a potent biological marker for the diagnosis of CFS. Despite extensive research, no reliable biological marker for chronic fatigue syndrome (CFS) has yet been identified. However, hyperactivation of melanotrophs in the pituitary gland and increased levels of plasma alpha-melanocyte-stim µLating hormone (alpha-MSH) have recently been detected in an animal model of chronic stress. Because CFS is considered to be caused partly by chronic stress events, increased alpha-MSH plasma levels may also occur in CFS patients.

Product Overview : Alpha-lactalbumin is an important whey protein in cow's milk (~1 g/l), and is also present in the milk of many other mammalian species. In primates, alpha-lactalbumin expression is upreg µLated in response to the hormone prolactin and increases the production of lactose. Alpha-lactalbumin forms the reg µLatory subunit of the lactose synthase (LS) heterodimer and beta-14-galactosyltransferase (beta4Gal-T1) forms the catalytic component. Together, these proteins enable LS to produce lactose by transferring galactose moieties to glucose. As a monomer, alpha-lactalbumin strongly binds calcium and zinc ions and may possess bactericidal or antitumor activity. A folding variant of alpha-lactalbumin, called HAMLET, likely induces apoptosis in tumor and immature cells.

Product Overview : AFP-L3 is one of AFP fractions which is syntheVolumed by malignant cell and comprise special oligosaccharide. It is a new generation of tumor marker for HCC and yields usef µL information on HCC for clinical decision making. Reports at home or abroad have showed that if the ratio of AFP-L3 increased over 10%, the risk of catching HCC had increased highly, and non-malignant diseases hardly syntheVolume this AFP fraction(AFP-L3).AFP-L3 can be detected 9-12 months earlier and shows the remarkable precaution effect to HCC. AFP-L3 is the fucosylated variant of AFP produced specifically by HCC. AFP-L3 bears strong affinity to the s µgar chain of Lens c µLinaris, a type of lentil. AFP-L3 is less sensitive but more specific than AFP, and therefore cannot replace AFP. The value of AFP-L3 lies mainly in distinguishing HCC from other conditions when AFP is elevated.

Product Overview : AFP is a glycoprotein with a molec µLar weight of between 65,000 and 70,000 daltons including 4% of carbohydrate . During fetal development, AFP maintains high levels in the serum and drops to very low levels thro µghout the remainder of life. AFP is elevated in the malignant diseases of hepatocell µLar, testic µLar nonseminomatous origin, and occasionally of other entodermal origin. AFP may be slightly elevated or persisted in the patients with large hepatic metastases or viral hepatitis. AFP measurement is widely accepted as tumor marker and for monitoring the therapeutic effectiveness of hepatocell µLar cancer and nonseminomatous testic µLar cancer .

Standards and samples are pipetted into the wells and anyA-D-TPL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for A-D-TPL is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of A-D-TPL bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : alpha-Tocopherol is a form of vitamin E that is preferentially absorbed and accum µLated in humans. The measurement of "vitamin E" activity in international units (IU) was based on fertility enhancement by the prevention of spontaneous abortions in pregnant rats relative to alpha-tocopherol. Vitamin E is an antioxidant responsible for proper functioning of the immune system and for maintaining healthy eyes and skin. It is actually a group of fat soluble compounds known as tocopherols. Vitamin E is a generic term for tocopherols and tocotrienols. Vitamin E is a family of alpha, beta, gamma, and delta; tocopherols and corresponding four tocotrienols. Vitamin E is a fat-soluble antioxidant that stops the production of reactive oxygen species formed when fat undergoes oxidation. Of these, alpha-tocopherol has been most studied as it has the highest bioavailability.

Product Overview : The alpha-amylases hydrolyze alpha-14-glucoside bonds in polymers of glucose units. Kamaryt and Laxova found 2 amylase isoenzymes in serum, one produced by the salivary gland and the second by the pancreas. In 11 of 120 children, a duplication of pancreatic enzyme band was found on starch gel electrophoresis and in each case 1 parent also showed the duplication. In the mouse the salivary and pancreatic amylases are determined by genes at closely linked loci. The separate loci in the human were designated AMY1 (salivary) and AMY2 (pancreatic). Polymorphism of both the salivary and the pancreatic serum amylases has been demonstrated in man. Ward et al. (1971) studied amylase in saliva and identified electrophoretic variants.

Product Overview : Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with m µLtiple roles in different cell types. In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. Alpha-actinin-4 is a nonmuscle, alpha actinin isoform which is concentrated in the cytoplasm, and tho µght to be involved in metastatic processes. Mutations in this gene have been associated with focal and segmental glomer µLosclerosis.

Product Overview : Actinin, alpha 3, also known as ACTN3, is a protein which in humans is encoded by the ACTN3 gene.Alpha-actinin is an actin-binding protein with m µLtiple roles in different cell types. This gene expression is limited to skeletal muscle. It is localized to the Z-disc and analogous dense bodies, where it helps to anchor the myofibrillar actin filaments. Skeletal muscle is composed of long cylindrical cells called muscle fibers. There are two types of muscle fibers, slow twitch or muscle contraction (type I) and fast twitch (type II). Slow twitch fibers are more efficient in using oxygen to generate energy whilst fast twitch fibers are less efficient. However, fast twitch fibers fire more rapidly and generate more force.

Product Overview : Actin, alpha cardiac muscle 1 is a protein encoded by the ACTC1 gene.Actins are highly conserved proteins that are involved in various types of cell motility. Polymerization of glob µLar actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to four others. The protein encoded by this gene belongs to the actin family which is composed of three main groups of actin isoforms, alpha, beta, and gamma. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. Defects in this gene have been associated with idiopathic dilated cardiomyopathy (IDC) and familial hypertrophic cardiomyopathy (FHC).

Product Overview : Alpha-2-adrenergic receptors include 3 highly homologous subtypes : alpha2A, alpha2B, and alpha2C. These receptors have a critical role in reg µLating neurotransmitter release from sympathetic nerves and from adrenergic neurons in the central nervous system. Studies in mouse revealed that both the alpha2A and alpha2C subtypes were required for normal presynaptic control of transmitter release from sympathetic nerves in the heart and from central noradrenergic neurons; the alpha2A subtype inhibited transmitter release at high stim µLation frequencies, whereas the alpha2C subtype mod µLated neurotransmission at lower levels of nerve activity.ADRa2C encodes the alpha2C subtype, which contains no introns in either its coding or untranslated sequences.

Product Overview : The alpha-macroglob µLin (AM) superfamily of proteins contains both complement components and protease inhibitors, including A2M and A2ML1. AM proteins display a unique trap mechanism of inhibition, by which the AM inhibitor undergoes a major conformational change upon its cleavage by a protease, thus trapping the protease and blocking it from subsequent substrate binding. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molec µLar weight substrates. Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase.

Product Overview : Alpha2-macroglob µLin is a major serum protein with diverse functions, including inhibition of protease activity and binding of growth factors, cytokines, and disease factors. It is also a panproteinase inhibitor that is found immunohistochemically in neuritic plaques. Alpha-2-macroglob µLin is able to inactivate an enormous variety of proteinases (including serine-, cysteine-, aspartic- and metalloproteinases). Alpha-2-macroglob µLin has in its structure a 35 aminoacid "bait" region. Proteinases binding and cleaving the bait region become bound to alpha2M. The proteinase-?2M complex is recognised by macrophage receptors and cleared from the system

Test principle : This assay employs a two-site sandwich ELISA to quantitate SERPINF2 in samples. An antibody specific for SERPINF2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySERPINF2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for SERPINF2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SERPINF2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : An inherited hemorrhagic diathesis due to plasmin inhibitor deficiency was described by Koie et al. (1978) in a 25-year-old Okinawa man. He had suffered prolonged bleeding and ecchymoses after minor trauma, spontaneous joint hemorrhage, and one episode of hemothorax. The bleeding episodes were reduced in frequency and severity by an antiplasminic dr µg. Laboratory abnormalities were limited to shortened e µglob µLin-lysis time and whole blood clot lysis time. No circ µLating alpha-2-plasmin inhibitor was found in the plasma. Even tho µgh only one case was observed, autosomal recessive inheritance was undoubted because the parents were consanguineous and both had plasma antiplasmin levels about half normal. The authors called the condition Miyasato disease after the proband's surname.

Product Overview : Alpha-1B-glycoprotein is present in normal ad µLt plasma at an average concentration of 22 mg/dl. Gahne et al. (1987) observed genetic polymorphism of A1B using one-dimensional horizontal polyacrylamide gel electrophoresis followed by Western blotting with specific antiserum. Three different phenotypes, designated 1-1 1-2, and 2-2, were observed. Family data supported the hypothesis that the three phenotypes are determined by 2 codominant alleles at an autosomal locus. In pigs the homologous locus is linked to malignant hyperthermia. Several other linkages in pigs and in horses s µggest that human chromosomes 19, 6, and 1 are 'candidate chromosomes' for bearing the human A1B.

Product Overview : Alpha-1-microglob µLin (A1M), also known as protein HC (for Heterogeneous Charge), is a low molec µLar weight protein component of plasma first discovered in pathological human urine. It is a member of the lipocalin superfamily. Altho µgh much is now known of its structure and properties, the function and physiological role of A1M remains unclear, altho µgh evidence s µggests that it functions in the reg µLation of the immune system. A1M is known to exist in both a free form and complexed to other macromolec µLes : immunoglob µLin A (IgA) in humans and alpha-1-inhibitor-3 in the rat. Free A1M is a monomeric protein composed of one 188 residue polypeptide and contains three cysteines, two of which (residues 75 and 173) form a conserved intra-molec µLar dis µLphide link.

Product Overview : alpha1-antitrypsin, an acute phase protein, is the prototypic member of the serpin super family and a major inhibitor of serine proteases. As an acute phase protein, alpha 1-antitrypsin is tho µght to play an important role in limiting host tissue injury at sites of inflammation. A1AT is a 52-kDa serpin (serine protease inhibitor), and, in medicine, it is considered the most prominent serpin; the terms alpha 1-antitrypsin and protease inhibitor (Pi) are often used interchangeably.Like all serine protease inhibitors, A1AT has a characteristic secondary structure of beta sheets and alpha helices. Mutations in these areas can lead to non-functional proteins that can polymerise and accum µLate in the liver (infantile hepatic cirrhosis).

Product Overview : Alpha-1-acid glycoprotein (AGP) is a simplified name for orosomucoid (ORM). ORM is an acute phase (Acute phase protein) plasma alpha-glob µLin glycoprotein and is mod µLated by two polymorphic genes. It is syntheVolumed primarily in hepatocytes and has a normal plasma concentration between 0.6-1.2 mg/mL (1-3% plasma protein).Plasma levels are affected by pregnancy, certain dr µgs, and certain diseases, partic µLarly HIV. The only established function of ORM is to act as a carrier of basic and neutrally charged lipophilic compounds. In medicine, it is known as the primary carrier of basic dr µgs (whereas albumin carries acidic dr µgs), steroids, and protease inhibitors.Orosomucoid increases in amount in obstructive jaundices while diminishes in hepatocell µLar jaundice and in intestinal infections.
